Clinical observation of efficacy of ASTRAPURE injection in prevention and treatment of neuroto-xicity induced by oxaliplatin-containing chemotherapy regimen
Objective: To observe the efficacy of ASTRAPURE injection in prevention and treatment of neurotoxicity induced by oxaliplatin-containing chemotherapy regimen.
Methods:From September 2006 to September 2008, 40 patients with stomach or large intestinal cancer were enrolled in the study. The patients comprised 23 men and 17 women with age of 32-75 years (mean age 60 years) and were randomly divided into two groups: The test group and the control group (20 cases in each group). All patients were treated with one cycle of a oxaliplatin-containing chemotherapy regimen, which comprised oxaliplatin 130 mg/m2 on day 1, fluorouracil 0.5 g on days 1-5, and calcium foliate 0.2 g on days 1-5; in the test group, 30 ml of ASTRAPURE injection was added to the regimen on days 1-7. The manifestation of peripheral neurotoxic reactions was observed and nerve growth factor levels were measured.
Results: The situation of neurotoxicity occurring was as follows: in the control group, 2 patients had grade 0 toxicity, 10 had grade 1 toxicity, 6 had grade 2 toxicity, and 2 had grade 3 toxicity; in the test group, 14 patients had grade 0 toxicity and 6 had grade 1 toxicity. The incidence rate of neurotoxicity in the test and control groups was 30% and 90%, respectively. The difference was statistically significant (P<0.01). In the test and control groups, the nerve growth factor levels were (167±10)ng/ml and (204±19)ng/ml before chemotherapy as well as (152±8)ng/ml and (133±12)ng/ml 2 days after chemotherapy, respectively. In the control group, the nerve growth factor levels were markedly decreased 2 days after chemotherapy compared with before chemotherapy. The difference between the two groups was statistically significant (P<0.01).
Conclusion: ASTRAPURE injection has some degree of efficacy in prevention and treatment of neurotoxicity induced by oxaliplatin-containing chemotherapy regimen.

Study of ASTRAPURE injection on Proliferation and Expression of Vascular Cell Adhesion Mokcule-1 and E-selectin in Endothelial Cells
Objective: To investigate the effect of ASTRAPURE injection on proliferation and expression of vascular cell adhesion molecule and E-selectin in human umbilical vein endothelial cells(HUVEC).To study the mechanism of ASTRAPURE injection on regulation of hematopoiesis.
Methods: (1) Human umbilical vein endothelial cells culture model was founded,Endothelial cells was identified by immunohistochemical staining and transmission electron microscope (2)Treating HUVEC with various concentration of ASTRAPURE injection (0ug/ml,4ug/ml,40ug/ml,400μg/ml,4000ug/ml),Analyzed the proliferation of HUVEC by MTT; Analyzed the proliferation phase of HUVEC using flow cytometry; Measure the expression of VCAM and E-selectin by ELISA.
Results: (1) The results of immunohistochemical staining on correlation antigens of VIII factor and Weible-Paladabody in the endothelial cells was observed by using transmission electron microscope.demonstrat that we established the human umbilical vein endothelial cells culture model.. (2)Every groups has different influences on cell growth(Time:P=0.000<0.05;Concentration:P<0.05).(3) There was significant change was found between the experiment group and the control group on the proliferation phase of HUVEC(P<0.05).The number of cells in S phase and G2/M phase were increased, significant(P<0.05). The number of cells in G0/G1phase were reduced . (4) Endothelial cells were cultured in vitro can secret VCAM-1 no stimulation . Different ASTRAPURE injection and TNF can promoted the secretion of VCAM-1 from endothelial cells .400μg/ml ASTRAPURE injection is the most proper concentration. (P<0.05) ,meanwhile, ASTRAPURE injection can promoted the roles of TNF (P<0.05).(5)Endothelial cells were cultured in vitro can not secret E-selectin no stimulation. ASTRAPURE injection can not promoted the secretion of E-selectin from endothelial cells directly. Endothelial cells can been activated by TNF , ASTRAPURE injection can promoted the secretion of E-selectin from activated endothelial cells(P<0.05).
Conclusions: This research suggest that , ASTRAPURE injection can support hematopoiesis through endothelial cells and adhesion molecule , ASTRAPURE injection may promote the homing of hematopoietic stem/progenitor cells. Theory basis was afforded for the clinical application of ASTRAPURE injection through this research.

Study of the inhibitory effect of ASTRAPURE Injection on experimental cardiac hypertrophy in rats

Objective: To investigate the inhibitory effect of ASTRAPURE Injection on experimental cardiac hypertorphy in rats.
Methods: 42 male SD rats were divided into 4 groups: age machthed control, model, ASTRAPURE Injection group and Captopril group. cardiac hypertrophy was induced by suprarenal abdominal aorta constriction. The drugs were administered for 8 weeks from 4 weeks after surgery at dosage of ASTRAPURE Injection 10g/kg(i.p) and Captopril 50mg/kg(p.o). Blood pressure (BP) was measured 4 times during the period of experiment, and hemodynamics, cardiac index, cell size of left ventricular myocardial and total protein of left ventricular tissue were detected for 8 weeks after treatment with drugs.
Results: 1. The BP of the aorta binded rats kept increase in comparison with those of the normal and the age matched control rats, and ASTRAPURE Injection showed no significant effect on it. 2. After 8 weeks treatment with ASTRAPURE Injection, the +dp/dt max was increased and t dp/dt was shortened, which indicated that the ability of heart contraction was enhanced. 3. Left ventricular index (left ventricular weight/body weight) was notably decreased by treatment with ASTRAPURE Injection. 4. The cell size of left ventricular myocardial and total protein of left ventricular showed lightly reduce by ASTRAPURE Injection treatment, but there is no significant difference compared with those of model rats.
Conclusion: ASTRAPURE Injection can improve the abnormal cardiac function and may contribute to the prevention of the development of left ventricular hypertrophy induced by pressure overload.

Effects of collagen matrices by covalent incorporation of heparin and loading with ASTRAPURE injection on anagenetic blood vessel
Objective: To investigate effects of collagen matrices by covalent incorporation of heparin and loading with ASTRAPURE injection on anagenetic capillaries, we established the chick chorioallantois model. The collagen matrices by covalent incorporation of heparin and loading with ASTRAPURE injection were placed and then the eggs were continuously incubated for 3 days. The number of capillaries in the vicinity of samples, the hemoglobin content inside the samples, the dry weight and the macroscopic observation were evaluated. We found the heparinized matrices had comparable angiogenic effects. The number of capillaries, the hemoglobin content, the expression of CD34 increased remarkably (P < 0.01). So we concluded that ASTRAPURE injection might be considered as an alternative or addition agent to promote the acidification of capillaries.

Study of the Influence of ASTRAPURE injection on the Apoptosis of Primary Cultured Neonate Rat Cerebral Cortical Neurons Caused by Hypoxia in Vitro
Objective: To investigate the protective mechanism of ASTRAPURE injection on the apoptosis of primary cultured cerebral cortical neurons caused by hypoxia.
Methods: Using the primary culure of cerebral neurons of postnatal rats in free-serum with neurobasal medium supplied with 2% B27 supplement,trypan blue exclusion,Hypoxic culture of neurons,Hoechst 33342 staining and immunocytochemistry.
Results: From 10mg/ml to 100mg/ml, ASTRAPURE injection evidently dropped the apoptosis rate of cerebral cortical neurons induced by hypoxia and increased Bcl-2 protein expression and decreased Bax protein expression in the cerebral cortical neurons induced by hypoxia,improved the ratio of Bcl-2/Bax.
Conclusion: The ASTRAPURE injection is able to prevent from the apoptosis of hypoxic neurons in primary cultured cerebral cortical neurons. The effect of anti-apoptosis is through up-regulation of Bcl-2 protein expression and down-regulation of Bax-2 protein expression.
